What it is

SNAP-8 is a synthetic eight-amino-acid peptide derived from the N-terminal sequence of SNAP-25 (synaptosomal-associated protein 25), a component of the SNARE complex involved in vesicle-mediated neurotransmitter release. It was developed by Lipotec S.A. (Barcelona, Spain) as a topical cosmetic active under the INCI name Acetyl Octapeptide-3, and it is classified in most markets — including the EU and the United States — as a cosmetic ingredient, not a pharmaceutical drug. SNAP-8 is structurally related to Argireline (acetyl hexapeptide-3/acetyl hexapeptide-8), which targets the same SNAP-25 binding site using a six-amino-acid fragment. SNAP-8 extends that chain by two residues, and its developer claims greater potency as a result, though independent head-to-head data are limited. Both peptides are marketed as topical alternatives to injectable botulinum toxin, though the analogy is mechanistic rather than clinical: the amounts and tissue depths achievable through topical application differ fundamentally from an intramuscular injection. The evidence base for SNAP-8 consists predominantly of in-vitro mechanistic work and small manufacturer-sponsored clinical studies. Independent, peer-reviewed, adequately powered trials are lacking. The most commonly cited figure — up to 63% reduction in wrinkle depth over 28 days — comes from manufacturer-funded testing. A split-face study using a cream containing 3% of a 0.05% Acetyl Octapeptide-3 solution applied twice daily for 28 days found wrinkle depth lowered by up to 38%, with an average decrease of 7.1%. These results should be interpreted cautiously given the small sample sizes and industry sponsorship typical of cosmeceutical ingredient research. A key biological uncertainty is skin penetration. Related data on acetyl hexapeptide-8 — the closest well-studied analogue — show that less than 0.2% of topically applied peptide penetrates the stratum corneum after 24 hours, and whether any fraction reaches the neuromuscular junction in biologically meaningful concentrations remains unconfirmed. If wrinkle improvements are observed in humans, epidermal-level effects (hydration, surface texture) may contribute alongside the proposed neuromuscular mechanism. SNAP-8 is used exclusively as a topical cosmetic ingredient and should not be reconstituted for injection.
